This laboratory exercise is designed for the college laboratory. The chemicals are hazardous, proper safety precautions and waste disposal methods are to be used (gloves, goggles and garbage). Discuss safety precautions and waste disposal for this lab with your chemical safety officer.

The three primary reagents used in this activity can be purchased through Sigma Aldrich http://www.sigmaaldrich.com/united-states.html).

DTNB, 5,5′-Dithiobis(2-nitrobenzoic acid), 6,6′-Dinitro-3,3′-dithiodibenzoic acid, Ellmann’s Reagent
(CAS# 69-78-3)
D8130-1G, D8130-5G, D218200-1G, or D218200-5G

Tacrine, 1,2,3,4-tetrahydroacridin-9-amine (CAS# 206658-92-6)
A79922-1G, A79922-5G, A3773-1G, or A3773-5G

Acetylthiocholine iodide (CAS# 1866-15-5)
A5751-1G, A5751-5G, 01480-1G, 01480-5G

Purchase only the amount needed of these three reagents to perform the current. For safety reasons, do not purchase excessive amounts of these compounds. Dispose of excess solutions and chemicals in the manner prescribed by your chemical safety officer.

Materials for the phosphate buffer, sodium phosphate monobasic and sodium phosphate dibasic can be obtained from most biological chemical supply houses such as Flinn Scientific, Ward’s Natural Science, Daigger Lab Equipment & Supplies, etc.
